3D-printed CBD suppositories: New approaches in the treatment of epilepsy

Researchers have developed an innovative 3D-printed suppository that effectively releases cannabidiol (CBD) for the treatment of epilepsy. The novel approach combines advanced 3D printing technology with molding techniques to create a hollow structure that is both functional and safe. The suppository consists of an outer shell containing CBD and an inner spring structure to support the shape.

A central goal of the development was to overcome the disadvantages of conventional oral CBD drugs, in particular the so-called first-pass effect. This effect significantly reduces the bioavailability of active ingredients, as a large proportion of the drug is broken down during passage through the liver. The hollow design of the suppository makes it possible to efficiently hold the CBD medication at the site of action. This makes it particularly suitable for children and people with digestive problems.

Tests have shown that the suppository releases CBD slowly over a period of five hours. Both in vitro and in vivo studies proved the safety of the system. In experiments with rats suffering from epilepsy, the treatment was able to reduce brain damage and inflammation. In addition, an improvement in the composition of the intestinal microbiota was observed, indicating potentially positive side effects.

The researchers used fused deposition modeling (FDM) with thermoplastic polyurethane filaments to produce the feather-like structure of the suppository. The outer shell was produced using a 3D-printed metal molded part containing a mixture of CBD, polyvinyl alcohol and polyethylene glycol.

Details of this research can be found in the paper “3D-printed cannabidiol hollow suppositories for treatment of epilepsy”.
